TRYSIN PURIFICATION BY AFFINITY CHROMATOGRAPHY AND SDS –PAGE TRYPSIN VISUALISATION
INTRODUCTION
Trypsin is among other three key digestive proteinases –pepsin and chymotrypsin-whereby
their main role is to break down complex protein molecules in to their simple forms; the amino
acids. For laboratory purpose trypsin is isolated from crude pancreas extract.
Affinity chromatography is a molecular used in separation and purification of generally all
biomolecules with regards of its chemical structure and biological function .Sepharose is maostly
used matrix where immobilization of ligand molecules are done. The ligand to be used should
have high affinity to the substrate- have specific and reversible binding sites and have chemically
modifiable group.
