(Ligated Vibrio fischeri chromosomal DNA and pGEM plasmid vector with Sal I, and transformed E.coli cells with recombinant plasmids. And then, checked if the cloning was successful. Blue colonies indicate that it contains only the plasmid, and white colonies indicate that the plasmid and inserts are sucessfully transformed into the E.coli, and if plasmid contain lux operon, it will glow.) 1. Why there are more blue colonies than white colonies on the plates? (%of white colonies were 0~11%) 2. Why so few of the white colonies glowing?
