Microbiology
For this task, complete the Aseptic Technique and Culturing Microbes LabPaq experiment. Next, complete the microbial identification experiments listed below. Then, write a lab report discussing the results of these experiments. The instructions for completing the LabPaq experiment are located at the “LabArchives” website found in the web links section below.
Microbial identification experiments:
• Biochemical Testing for Microbial Identification-Methyl Red, Voges-Proskauer, and Catalase LabPaq experiment using Escherichia coli and Staphylococcus epidermidis
• Biochemical Testing for Microbial Identification-Carbohydrate Fermentation LabPaq experiment using Saccharomyces cerevisiae and Staphylococcus epidermidis
• Bacterial Identification through Functional Media-Motility Testing LabPaq experiment usingEscherichia coli and Staphylococcus epidermidis
• Environmental Influences on Growth-Salt Tolerance Testing LabPaq experiment usingSaccharomyces cerevisiae and Staphylococcus epidermidis
Note: You will need the Staphylococcus epidermidis culture for the next task, so do not discard it.
Your lab report should be written in complete sentences and in paragraph form. Your report should include the following sections: introduction, materials and methods, results, and discussion. The information required for each section is outlined below.
Requirements:
A. Write a lab report about the Aseptic Technique and Culturing Microbes LabPaq experiment and the microbial identification experiments in which you do the following:
1. Discuss the importance of using proper aseptic technique when working with microorganisms.
a. Discuss the importance of generating a pure culture.
Note: The discussions from parts A1 and A1a belong in the introduction of the lab report.
2. Explain how the following experiments were performed:
a. Biochemical Testing for Microbial Identification-Methyl Red, Voges-Proskauer, and Catalase LabPaq experiment
b. Biochemical Testing for Microbial Identification-Carbohydrate Fermentation LabPaq experiment
c. Bacterial Identification through Functional Media-Motility Testing LabPaq experiment
d. Environmental Influences on Growth-Salt Tolerance Testing LabPaq experiment
Note: The explanations from parts A2a-d belong in the materials and methods section of the lab report. Your explanations should focus on organisms used, media used, the time of incubation, and any reagents that were added.
3. Explain the results obtained for each microbial identification experiment performed on the following organisms:
a. Escherichia coli
b. Staphylococcus epidermidis
c. Saccharomyces cerevisiae
Note: The explanations from part A3a-c belong in the results section of the lab report. You may use the attached “Data Tables” to record your results to help you organize your response.
4. Describe the characteristics of each of the following organisms based on the results of allthe microbial identification experiments that you performed:
a. Escherichia coli
b. Staphylococcus epidermidis
c. Saccharomyces cerevisiae
Note: The descriptions from part A4a-c belong in the discussion section of the lab report.
Note: You will only discuss the biochemical characteristics that you observed. For example, Escherichia coli was not tested for salt tolerance, so a discussion of the salt tolerance of Escherichia coli would not be included.
B. Acknowledge sources, using in-text citations and references, for content that is quoted, paraphrased, or summarized.
Note: When using sources to support ideas and elements in a paper or project, the submission MUST include in-text citations with a corresponding reference list for any direct quotes or paraphrasing. It is not necessary to list sources that were consulted if they have not been quoted or paraphrased in the text of the paper or project.
